Effects of sildenafil in combination with angiotensin-converting enzyme inhibitor on limiting infarct expansion in a porcine model of acute myocardial infarction

      We investigated whether sildenafil could limit infarct expansion in a porcine model of acute myocardial infarction (MI). Twenty-four pigs were assigned to group 1 (control), group 2 (perindopril 2 mg daily), group 3 (sildenafil 50 mg daily), or group 4 (perindopril 2 mg plus sildenafil 50 mg daily). Infarct size was assessed by MDCT scan at 3–5 days and 30 days. Infarct size reduction showed incremental values across the groups (−7.7±5.1%, −13.8±8.2%, −15.3±8.3%, −17.8±5.0, p=0.086) with greatest difference between group 4 and group 1 (p=0.032). Sildenafil in conjunction with an ACE inhibitor may be effective in limiting infarct expansion.
